The Transcreener ADP2 Assay Detection without Coupling Enzymes

When you are on the search for a kinase assay kit that offers a single addition assay method and a direct measure of ADP in ATPase or kinase reactions, Transcreener ADP2 should be a top choice. This assay method was first introduced in 2006 as fluorescence polarization assay. However, over the years, it has been improved by the introduction of a higher affinity antibody that leads to increased sensitivity.

Instrument Compatibility

These kinase activity assays have been known and proven to offer quality screening for various targets including lipid, protein, and carbohydrate kinases, as well as carboxylases and chaperonin ATPases. At this time, rather than being used only for FP detection, it is also compatible with fluorescence intensity and time-resolved Forster resonance energy transfer. As such, the robustness and sensitivity with each of these is higher than with traditional assay methods.

The Real Difference

When using any of the three Transcreener kinase assay kits, you can expect Z’ values of better than 0.7 at10% or lower ATP conversion under initial velocity concentrations. The same was found with EC50 values and inhibitor profiles for PKA. This shows the equivalence associated with the assays for profiling of inhibitors.

Direct Measuring of Enzymatically Generated ADP

Transcreener is shown to offer the most direct and straightforward method for detecting ADP. ADP binds to a tracer, which creates a change in its normal fluorescence properties. This applies with TR-FRET, fluorescence intensity, and fluorescence polarization. On the other hand, the methods that utilize coupling enzymes to convert ADP into a different product that can be detected require are more laboriousIn coupled assays, eporter and coupling enzymes are used in order to create a signal. The problem is that each enzyme is a separate target for the screening compounds. This can lead to missing a hit or getting false positive. In addition, some methods require the use of extra liquid, incubation steps, or both. This adds time to the assay automation process.
